The difference between the two

NAD+ is a coenzyme found in every cell, where it acts as an electron carrier in energy metabolism. NMN, nicotinamide mononucleotide, is the immediate precursor to NAD+ in the salvage pathway: the enzyme NMNAT joins NMN to an ATP molecule and releases NAD+. So one formula supplies the coenzyme and the other supplies the precursor. Both act on the same pathway and either stands on its own. We do not recommend taking both, or taking them in any particular order.

What is established, and what is not

The cell biology above is textbook material and is not in dispute. What a supplement does in a person is a separate and much less settled question. A 2025 review in Nature Metabolism found the human trial evidence for NAD+ precursors limited, the data on human tissue sparse, and even the claim that NAD+ declines with age less consistent in people than in rodents.
